Human Osteosarcoma with mEmerald-PMP
In 1964, J. Ponten and E. Saksela derived the U-2 OS line from a moderately differentiated sarcoma of the tibia of a 15-year-old Caucasian girl. Viruses were not detected during co-cultivation with WI-38 cells or in CF tests against SV40, RSV, or adenoviruses. The U-2 OS cells are positive for insulin-like growth factor I (IGF-I) and insulin-like growth factor II (ICF II) receptors. The human osteosarcoma cells (U-2 line) appearing in the selected digital video sequence are visualized expressing mEmerald fused to peroxisomal membrane protein (PMP), highlighting peroxisomes.
A variation of the enhanced Aequorea derivative EGFP, mEmerald is a green fluorescent protein named for the gemstone that exhibits a similar hue. In addition to the S65T and F64L mutations found in EGFP, mEmerald benefits from four additional point mutations that promote improved brightness and photostability. Excitation and emission maxima of mEmerald occur at 487 and 509 nanometers, respectively.
