Rabbit Kidney Cells with mEmerald-MAP4-N and mApple-H2B
Rabbit kidney epithelial cells (RK-13 line) appear in this digital video sequence expressing mApple fused to H2B and mEmerald fused to MAP4. H2B and MAP4 are markers of cell nuclei and microtubules, respectively. H2B is a highly conserved histone protein that forms a complex with other histones to form the core of nucleosomes. MAP4 promotes microtubule assembly, and has been proved to offset interphase microtubule catastrophe promotion damage. Cyclin B, which targets cell division cycle 2(CDC2 kinase to microtubules, was experimentally discovered to interact with MAP4.
One of the red members of the mFruit series of fluorescent proteins, mApple is a monomeric protein developed from the directed evolution of mRFP1. Excitation of mApple peaks at 568 nanometers and emission peaks at 592 nanometers. mEmerald is an improved green variant of EGFP, which is an enhanced version of the green fluorescent protein initially isolated from the jellyfish Aequorea victoria. Excitation and emission maxima of mEmerald occur at 487 nanometers and 509 nanometers, respectively.
