Monkey Kidney Cells with mEmerald-PMP and mApple-EB3
EB3 often finds use in microscopy applications as a marker of microtubules. The protein is linked with the cytoplasmic microtubule network, is particularly upregulated upon myogenic differentiation, and knockdown of EB3 averts myoblast elongation and myotube synthesis. It is essential for the microtubule dynamics regulation and capture of microtubules at the cell cortex, a crucial step during myogenic differentiation and possibly a wide-reaching mechanism in polarized cell elongation. A chimera of EB3 and mApple was employed to visualize the microtubules present in the African green monkey kidney cells (CV-1 line) shown in this digital videos sequence. mApple is a red fluorescent protein that matures rapidly and exhibits excitation and emission maxima at 568 and 592 nanometers, respectively.
The featured fibroblast cells were also tagged with a green fluorescent protein, mEmerald, fused to a peroxisomal membrane protein targeting signal. mEmerald is an EGFP variant with improved brightness and photostability. In addition to mutations specific to EGFP, mEmerald features four additional point mutations. Excitation and emission of the fluorescent protein peak at 487 nanometers and 509 nanometers.
